Incorporation of ZnO/bioactive glass nanoparticles into alginate/chitosan composite hydrogels for wound closure

To develop a proper skin wound dressing is a vital step in accelerating the wound closure process and enhancing the survivability of patients with serious skin injuries. Herein, a zinc-doped bioactive glass (ZBG)/succinyl chitosan (SCS)/oxidized alginate (OAL) composite hydrogels (Gel-ZBG) have been developed as the wound dressings to accelerate the wound closure. The Schiff-based linkages have been introduced into the composite hydrogels which provide a humid microenvironment for the proliferation of cells on wound sites. The amino groups from SCS and Zn ions released from ZBG made the excellent antibacterial properties to composite hydrogels that confirmed by the antibacterial tests in vitro. The Si and Ca ions can stimulate the fibroblast cells to develop essential growth factors for angiogenesis and wound closure processes. The epidermal growth factors (EGFs) embedded into the hydrogels can significantly improve cell proliferation and tissue remodeling in the wound bed. To evaluate the potential of hydrogels as skin wound dressings, cytotoxicity, bacteriostatic properties and wound closure of the organism have been investigated. The formation of granulation tissue, deposition of collagen and myofibril, the release of anti-inflammatory factors and angiogenesis have been evaluated to determine the healing mechanism using the obtained composite hydrogels as the wound dressings.
