A two-stage energy tuning strategy via salt and glycine programmed DNA-engineered crystals

摘要:为在室温下获得高质量DNA工程晶体,提出了一种先加入NaCl再加入甘氨酸(Gly)的两阶段能量调控策略。Gly的加入可精细平衡结晶的吸引和排斥能量。由能量而非温度诱导的状态转变对生物体系具有重要意义。考虑到DNA识别和聚电解质性质,该两阶段能量调控策略通过改变磷酸基电荷中和比例,利用盐和Gly精细调控DNA相互作用,从而在室温下构建高质量晶体。

Abstract:To obtain high-quality DNA-engineered crystals at room temperature, a two-stage energy tuning strategy by first adding NaCl and then glycine (Gly) is proposed. The addition of Gly can exquisitely balance the attraction and repulsion energies for crystallization. The state transition induced by energy rather than temperature is significant for a biosystem. Considering the properties of DNA recognition and polyelectrolyte, the two-stage energy tuning strategy is proposed to mediate DNA interactions finely via salt and Gly by changing the fraction of phosphate charge neutralization, enabling construction of high-quality crystals at room temperature.
