Niobium carbide doped ROS/temperature dual-responsive multifunctional hydrogel for facilitating MRSA-infected wound healing

The satisfactory healing of chronic wounds associated with multi-drug resistance (MDR) bacterial infection remains a challenge owing to the persistent inflammation and severe bacterial infection. Herein, we develop an antibacterial anti-oxidative Nb2C MXene hydrogel (FGDN) to alleviate excessive oxidative stress and eradicate bacterial infection for accelerating MRSA-infected wound healing. FGDN hydrogel were prepared by Schiff base reaction between Pluronic F127 aldehyde (FCHO), branched poly (glycerine-amino acid) (GEPL), PDA-modified Nb2C MXene nanosheets (Nb2C@PDA), and 3,3′-dithiodiazine (propionyl hydrazide) (DTPH). FGDN hydrogel exhibited outstanding injectability, conductivity, self-healing, anti-oxidation, tissue adhesion and rapid hemostatic ability. Additionally, FGDN hydrogel could efficiently eliminate excess reactive oxygen species and protect cells from oxidative stress damage in vitro. Meanwhile, FGDN hydrogel could promote cell proliferation with negligible toxicity. Besides, FGDN hydrogel displayed astonishing antibacterial capacity for MDR bacterial MRSA (antibacterial efficiency was about 100% in vitro) without the help of antibiotics. Most importantly, FGDN significantly accelerated MRSA-infected wound healing (wound closure rate: 97.30%) by quickly hemostasis, eradicating bacterial infection, anti-inflammatory, stimulating cell proliferation, angiogenesis, promoting granulation tissue generation, collagen deposition and re-epithelialization. The multifunctional FGDN hydrogel provides an ideal dressing for MRSA-infected wound healing/tissue regeneration.
