报告摘要:

研究DNA/RNA 与聚阳离子的静态和动态复合行为对于探讨聚电解质的相互作用机理,开发高效基因传递载体,以及研究原始细胞的动态模型都有重要意义。在静态复合中,我们重点探讨了DNA复合物的长程动力学行为,发现歧化是影响复合物动力学的主要因素。通过调控歧化速率,构筑了动力学控制的胶囊结构。在动态复合中,首先模拟血液流动引起的剪切作用,探讨了外加剪切力对DNA复合物稳定性的影响,并据此开发了具有肺靶向功能的siRNA传递载体。 接下来,利用电场来调控DNA与聚阳离子在非平衡态的复合行为。结果表明,二者形成的复合物发生了周期性的内分区化,尺寸和形状的动态涨落,混沌式的增长和融合等许多与生命活动类似的行为。在周期性的内分区化过程中还伴随着物质的运输,主要表现为聚阳离子含量的减少。我们用电场力学引起的聚电解质复合物的解散,继而引起的渗透压变化来解释内分区化行为。这些结果都表明在非平衡态下研究复合物粒子的动态行为对于开发原始细胞,探索生命起源具有指导意义。
