摘要:糖尿病慢性创面因长期炎症反应、细菌感染以及组织再生能力受损而难以愈合,仍是亟待解决的临床问题。本研究构建了一种活性氧响应型一氧化氮释放水凝胶体系,以三羟甲基氨基甲烷改性的透明质酸为基体,引入具有天然抗菌活性的 ε-聚赖氨酸,并结合功能化中空介孔杂化二氧化硅纳米颗粒,实现药物负载与刺激响应释放。该水凝胶可在温和条件下快速凝胶化,形成稳定三维网络结构,表现出良好的组织黏附性能、抗菌活性及促愈合潜能。通过结构与功能模块的理性设计,材料在氧化应激条件下可实现异维A酸的可控释放,并进一步触发按需 NO 生成。体内外实验表明,异维A酸与 NO 的释放均呈 ROS 水平依赖性且具可调控特征,从而显著促进创面闭合,增强胶原沉积与血管新生,并改善局部炎症微环境。该水凝胶体系具有良好的生物相容性与智能响应特性,提示其作为多功能创面敷料在糖尿病慢性创面高效治疗中具有潜在应用价值。
