Biodegradablepolymers(e.g.,polylactic-co-glycolicacid(PLGA》havebeenextensivelyinvestigatedforcancerdiagnosisandtherapy.NPssizeplaysakeyroleindegradation,circulationtime,drugrelease,biodistribution,clearance,uptakemechanismsandtoxicity.Narrowsizedistributiongivescontroloverthedoseandreleasebehaviorofthedrugsandyieldshighdrugencapsulationefficiencyandgoodbiocompatibilityc[l].Malignantmelanomarepresentsaformidablechallengeduetoitshighmortalityrateworldwide.However,aclinicaltrialofnab-paclitaxelshoweditshighresponserateinmetastaticmelanomac[2].Wewillpresentasimple,yetversatilestrategyforthefabricationofuniformbiodegradablepolymerNPswithcontrollablesizesbymembrane-extrusionemulsificationapproach.SizeandsizedistributionoftheNPscanbeeasilytunedbyvaryingexperimentalparameters.Moreover,hydrophobicdrugsandfunctionalinorganicNPscanbeeffectivelyandsimultaneouslyencapsulatedintothepolymerNPstoformmultifunctionalhybridNPs.Thesedrug-loadedNPsexhibithighencapsulationefficiencyanddrugloadingdensityaswellasexcellentdrugsustainedreleaseperformance.MelanomacellexperimentsindicatethatthePTX-IoadedhybridNPsproducedbythistechniquecouldbepotentiallyappliedasamultifunctionaldeliverysystemfordrugdelivery,bio-imaging,andtumortherapy,aswellasmalignantmelanomatherapy.
