Magneticironoxidenanoparticles(MIONs),owingtotheiradvantagessuchaslowtoxicityandbiocompatibility,areconsideredtobethemostfavorablecandidatesforbio-applicationsincludingmagneticresonanceimaging(MRI),magneticfluidhyperthermia(MFH),magneticseparationandimmobilizationofbiomoleculessuchasnucleicacidsandproteins,thedevelopmentofdrugdeliverysystemsforcontrolledreleaseofdrugsbiolabelingandmagneticsensors.[1]Inthisstudywehavedescribedasimpleonestepprotocolforthepreparationoffairlymonodisperseandhighlywater-solublemagneticironoxidenanoparticles(MIONs)throughco-precipitationmethodusinganovelmultifunctional,biocompatibleandwater-solublepolymerliganddodecanethiol-polymethacrylicacid(DDT-PMAA).DDT-PMAA,owingtoitsseveralintrinsicproperties,notonlyefficientlycontrolsthesizeoftheMIONsbutalsogivesthemexcellentwatersolubility,longtimestabilityagainstaggregationandoxidation,biocompatibilityandmultifunctionalsurfacerichinthioetherandcarboxylicacidgroups.Themolecularweightandconcentrationofthepolymerligandwereoptimizedtoproduceultrasmall(4.6+0.7nm)MIONswithhighmagnetization(50emug-l)Doxorubicin(DOX)throughitsaminegroupswasconjugatedtothecarboxylicacidgroupsofMIONsthroughelectrostaticandcovalentinteractionstoprepareDOX/MIONsandDOX-MIONsrespectively.NPsanddrugconjugatesshowedhighdrugloadingefficiency(upt060%)anddemonstratedhighercytotoxicitytowardsHepG2cancercellsduetotheirimprovedsolubilityandcellularuptakethanthefreedrug.ItwasobservedthatdrugloadingmodecansignificantlyaffectthedrugefficiencyandthisstudycanhelpintheoptimizationofdevelopmentoftheMIONsbaseddrugdeliveryformulations.MIONsstabilizedwithDDT-PMAAhaverichsurfacechemistrywhichcanbeexploitedfortheconjugationofavarietyoftherapeutic,targeting,andlabellingagentsforseveralbio-analyticalanddrugdeliveryapplications.
