Self-assemblednanostructuresfromdiblockcopolymersarebeingintensivelyinvestigatedfortheirpotentialdrugdeliveryapplications,especiallythesyntheticcopolypeptideandpolypeptidebasedhybridcopolymersbecauseoftheiruniquestructuralpropertiesandbiocompatibility.Recently,thepreparationofwell-definedpolypeptide,poly(γ-propargyl-L-glutamate)(PPLG)whichcontainspendantalkynegroupsonside-chain,hasbeendeveloped.Herein,wesynthesizedPoly(ethyleneglycol)-Poly(γ-propargyl-L-glutamate)(PEG-b-PPLG)blockcopolymersbythering-openingpolymerization(ROP)usingawater-solubleamine-terminatedpoly(ethyleneglycol)(PEG-NH2)astheinitiator,wherethePPLGblockwasmodifiedwithahydrophobic,lipophobic,fluorinatedsidegroupandasaturatedhydrocarbonbythiol-ynereactionofthealkynegroupswith1-mercapto-1H,1H,2H,2H-perfluorooctaneand1-octanethiol.Thefunctionalizedpolypeptidescanself-assembleintodisc-likestructuresinaqueousandorganicsolutionwithcompartmentalizedcoronasduetothedifferentsolubilitycharacterofthetwodisparateblocksandtheα-helicalconformationofthepolypeptideblock.
